# Do different doses of medroxyprogesterone affect mood in women on hormone replacement therapy?

Both doses caused mood symptoms, but the higher dose improved mood in women with a prior PMS history. This small randomized trial suggests lower doses do not help.

 Moderate study design

36 postmenopausal women in northern Sweden, some with a history of PMS

## What they did

A randomized double-blind crossover study of 36 postmenopausal women in northern Sweden. Women received 2 mg estradiol continuously and either 10 mg or 20 mg medroxyprogesterone acetate for 12 days of each cycle, for 5 cycles. Mood and physical symptoms were tracked daily.

## What they found

Physical symptoms did not differ between doses. Both groups had more negative mood with the 10 mg dose. Women with previous PMS had better positive mood with the 20 mg dose.

## What it means for you

In women on hormone replacement therapy, the higher dose of this progestin may be better for mood, especially if you have a PMS history. The study was small and included only postmenopausal women in Sweden.
